Difference between blood agar and chocolate agar culture medium

Blood agar and chocolate agar are the most famous and widely used culture media at present. However, many people do not know the difference between these two cultivation environments. In the next article, we intend to introduce these cultivation environments and their differences.

In the previous articles, the types of culture medium were explained; The uses of each of them are mentioned and a brief reference is made to the most famous cultivation environments. To check the difference between Blood Agar and Chocolate Agar culture media, it is necessary to get acquainted with each of these culture media separately. For this introduction, we will start with blood agar culture medium.

Difference between blood agar and chocolate agar culture medium

Blood agar culture medium

Blood agar is one of the types of enrichment culture media that is used for the growth of microorganisms that are hard to grow. Such microorganisms do not grow well in normal culture media. This culture medium is also used to detect the hemolytic properties of bacteria.

Blood agar contents

  • 0.5% Peptone
  • 0.3% beef extract
  • 1.5% agar
  • 0.5 sodium chloride (NaCl)
  • Distilled water
  • 5% sheep blood

*pH should be between 7.2 and 7.6.

Chocolate agar culture mediumChocolate agar culture medium

Chocolate agar culture medium is an enriched culture medium in which lysed blood globules (their membranes have been destroyed) are present. These lysed blood cells turn brown; That is why they are called chocolate agar. In fact, chocolate agar does not contain any chocolate.

The lysis of red blood cells causes the release of intracellular compounds such as hemoglobin, some enzymes and various factors. These factors help a lot in the growth of cells and bacteria.

The contents of chocolate agar

The contents of chocolate agar are the same as blood agar, and the only difference is that in chocolate agar, red blood cells have been lysed. (Their contents are spread in the culture medium.)

The difference between blood agar and chocolate agar

The difference between these two culture mediums is that the chocolate agar culture medium has been heated, as a result of this heat, its red blood cells have been lysed and their contents have been spread in the medium. These contents have a great effect on the growth of bacteria and cells.
